Development of Metronidazole Sustained-release Pellets

 

甲硝唑(Metronidazole)又名甲硝基羟乙唑、灭滴灵、灭滴唑。除用于抗滴虫和抗阿米巴原虫外,近年来,广泛地应用于抗厌氧菌感染。甲硝唑是通过抑制阿米巴原虫的氧化还原反应,使原虫的氮链发生断裂而抗阿米巴原虫的。它在抗菌过程中,硝基被还原成一种细胞毒,从而作用于细菌的DNA代谢过程,促使细菌死亡。最新的研究表明,甲硝唑对放疗有增敏作用,还可用于高脂血症等。现在市面上甲硝唑的口服剂型多为每日3次的普通剂型,由于它最常发生的不良反应为胃肠道反应,为了减少患者的给药频率,增加患者的顺应性,降低不良反应的发生率,本实验采用离心造粒粉末层级法制备甲硝唑微丸,并用丙烯酸树脂水分散体对其进行包衣,制备出甲硝唑缓释微丸。

 

Metronidazole is also known as fossyol and flagyl. In addition to being used for anti-trichomonas and anti-amebic protozoa, in recent years, it is widely used ianti-anaerobicn infections. Metronidazole fights against amebic protozoa by inhibiting the redox reaction of the amebic protozoa, and then causing the protozoan's nitrogen chain to rupture. During its antibacterial process, the nitro is reduced to a  cytotoxin, which acts on the DNA metabolism of bacteria to promote bacterial death. The latest research shows that metronidazole has a sensitizing effect on radiotherapy, and it can also be used for hyperlipidemia. Now the oral dosage forms of metronidazole on the market are mostly ordinary dosage forms taken by 3 times a day; because its most common adverse reaction is gastrointestinal reaction, in order to reduce the patient's frequency of administration, increase patient compliance and reduce adverse reactions, in this experiment, metronidazole pellets were prepared by centrifugal granulation powder gradation method and coated with aqueous dispersion of acrylic resin to prepare Metronidazole Sustained-release Pellets.
